Methods of use of Compound Microscope:
- Place the Microscope on table where sufficient light is available.
- Adjust Position And Height of the observer's chair comfortable for the observation. Check that the objective, eyepiece, stage and mirror is totally free from dust.
- Clean the Lens of eyepiece, Objective Lens with the xylol to remove harden oil.
- Pick up the condenser using the screw about 1 mm below the stage.
- Fully open the irris diaphragm.
- Adjust the light with Sufficient intensity by using low power objective lens.
- Place the slide with the object which is to be observe on stage and adjust exactly below objective lens.
- Position of the oil immersion lens in proper place when light magnification is required.
- Use the coarse adjustment screw to focus specimen on slide.
- Move the area of slide for easily visible material at the centre of field.
- Never use the fine adjustment screw until the specimen has been made visible and brought nearly into the focus with coarse adjustment. Use fine adjustment only to obtain the maintain exact focusing.
- Before using oil immersion lens, place a drop of immersion oil (Cidar Wood Oil) on the middle of the smear. The addition of excess oil is exactly avoided.
- Adjust the object first by using coarse adjustment screw and then make it very accurate by using fine adjustment screw.
- After Observation. remove the oil from the objective lens by using xylene is avoided because it may dissolve cementing substance used in objective lens.
Care for Using Microscope:
- The microscope is most important instrument in the microbiology laboratory.
- It should be used with care and maintain properly.
- It should always protected from dust and kept in closed container.
- it should be cleaned it interval.
- The oil immersion lens is cleaned with xylene after use to remove the oil.
